Does the weight of new tiles affect the roof?

It may do.  If you are replacing your roof like-for-like then there should be no problem. However, different tile materials do have different weights, so if you are changing the tile material you should check with a structural engineer to make sure the existing rafters are strong enough to bear the weight of the new tiles.
